    EFFECT OF DIFFERENT SEMEN EXTENDERS ON CAT EPIDIDYMAL SEMEN CRYOPRESERVATION

    BACKGROUND: Cryopreservation of spermatozoa is a biotechnology used for fertilization purposes and preservation of genetic material in various domestic species. OBJECTIVE: To determine the efficacy of two different commercial semen diluents in the cryopreservation of epididymal semen of domestic cats. MATERIALS AND METHODS: Twenty male cats aged between 1 - 3 years and weighing 2.5 - 4.5 kg were used in the study. The testicular tissues removed from the cats were immediately brought to the laboratory in physiological saline and the epididymal parts were trimmed in commercial semen extenders (INRA 96, Group I; OPTIXCELL, Group II). Diluted semen samples were cooled to 4 degrees C and filled into 0.25 mL straws. Semen samples were frozen in a programmable semen freezing device and then placed in a liquid nitrogen container at -196 C. Semen samples were thawed at 38 degrees C for 25 s. Thawed semen samples were evaluated in terms of motility and kinematic parameters using CASA. RESULTS: No statistical difference was found between the groups in terms of total motility, progressive motility, and velocity parameters at 4 degrees C. The rate of spermatozoa at slow speeds was found to be lower in group II. In addition, after freezing and thawing process, no statistical difference was observed between the groups in terms of motility, kinematics, and velocity parameters. CONCLUSION: Both commercial semen extenders can be used for cryopreservation of cat epididymal

    Investigation of changes in Hematological parameters and levels of Oxidative stress factors in castrated Cats

    This study was carried out to determine the effect of castration procedure on hemogram and leukogram values and the levels of oxidative stress factors in serum tissue in cats. Preoperative and postoperative 10th day hemogram/leukogram values and total oxidant/antioxidant capacities of 19 castrated cats were evaluated in the study. When the hemogram and leukogram values were examined, it was determined that there was a statistically significant increase in the amount of eosinophils (P<0.05). Although the increase in white blood cells (WBC), neutrophil and lymphocyte levels and the decrease in red blood cells (RBC) and hematocrit (HCT) levels were remarkable, it was not found to be statistically significant (P>0.05). When the levels of oxidative stress factors in the serum tissues of the cats were examined, it was determined that there was a decrease in the total antioxidant status (TAS) and an increase in the total oxidant status (TOS), but the difference in the results was not statistically significant (P>0.05). In conclusion, in this study, it was determined that the castration procedure in cats did not cause a significant difference in hemogram and leukogram parameters and the level of oxidative stress factors in serum tissue, except for the amount of eosinophils.

    Rock salt and boron supplementation: Effects on oxidative stress in blood and semen of rams

    This study aimed to investigate the effects of boron and rock salt minerals, which are interesting as antioxidant minerals on malondialdehyde (MDA) and some antioxidants; glutathione level (GSH), glutathione peroxidase (GSH-Px), superoxide dismutase (SOD), and catalase (CAT) enzyme activities in blood and sperm cells. For this purpose, a total of 36 rams were divided into six research groups, with six rams in each group. The research groups were designed as Control (Just 40 ml water), Salt (10 g rock salt/day), B20 (20 mg boric acid/day), B40 (40 mg boric acid/day), BT20 (20 mg boric acid + 10 g rock salt/day), BT40 (40 mg boric acid + 10 g rock salt/day). The additives were dissolved in 40 ml of water and administered orally with 50 ml syringes for 80 days. Semen and blood samples were taken from the rams at the end of the experiment. Salt added to the diet significantly increased MDA levels in blood and sperm cells. A significant decrease in plasma MDA levels were observed in the BT40 group and sperm cells in the B40 and BT40 groups. Salt and boric acid were found to be effective on GSH and antioxidant enzymes. In conclusion, it was found that boric acid added to the diet at a dose of 40 mg enhanced antioxidant metabolism and significantly reduced salt-induced lipid peroxidation. © (2025), (Hellenic Veterinary Medical Society).
