    Antioxidant effect of grape molasses in rat heart tissues

    Grape molasses, which is rich in mineral substances such as flavonoids, polyphenol, antioxidants, iron, calcium, phosphorus, potassium, and magnesium is a natural nutrient, that is concentrated with boiling without adding any additive. In this study, we aimed to investigate the effects of molasses and resveratrol diets on oxidative stress parameters in cardiac damage induced with 7,12-dimethylbenz(a)anthracene (DMBA). A total of 42 Wistar albino female rats were divided into six groups with seven in each. Control group was administered subcutaneous injection of 1 mL mixture including 20 mL sesame oil + 30 mL Dimethyl Sulphoxyde (DMSO); DMBA group received subcutaneous injection of 10 mg/Kg DMBA on the 0th and 7th days. DMBA + Molasses group received DMBA application and feed containing 20 % molasses; DMBA + Resveratrol group received subcutaneous injection of DMBA + 10 mg/Kg resveratrol a day. Molasses group received feed with 20 % molasses, and Resveratrol group received subcutaneous injection of 10 mg/Kg resveratrol a day. There was a statistically significant difference between the DMBA group and DMBA + Molasses, control, resveratrol and molasses groups in terms of nitric oxide activity. There was a statistically significant difference between the DMBA group and DMBA + Resveratrol, DMBA + Molasses, control, molasses and resveratrol groups in term of malondialdehyde (MDA) activity. There was a statistically significant difference between DMBA groups, and DMBA + Molasses, DMBA + Resveratrol, and control groups in terms of glutathione (GSH) activity. Based on these results, it could be said that grape molasses could provide protection against oxidative stress as resveratrol, decreasing the risk of damage by free radicals.

    Assessing lipoxin-mediated inflammatory responses in the second trimester of pregnancy among women with obesity: A comprehensive analysis

    Objective: This study aimed to explore the relationship between maternal plasma lipoxin A4 (LXA4) levels during the second trimester of pregnancy and certain proinflammatory molecules, such as interleukin-6 (IL-6) and tumor necrosis factor alpha (TNF-?), as well as the antiangiogenic factor vascular endothelial growth factor receptor 1 (VEGFR-1), in conjunction with obesity among pregnant women. Materials and Methods: A total of 30 pregnant women with obesity were compared with 30 pregnant women of normal weight, matched for both age and gestational week. Plasma samples were collected from all participants between the 18th and 28th weeks of pregnancy. The levels of LXA4, VEGFR-1, IL-6, and TNF-? were quantified using enzyme-linked immunosorbent assay. Results: Plasma levels of LXA4 were notably lower in pregnant women with obesity, whereas levels of TNF-? and VEGFR1 were significantly higher (p=0.041, p<0.001, and p<0.001, respectively). There was no significant difference in IL-6 levels between groups (p=0.072). The binary logistic regression model revealed significant associations between obesity and the examined inflammatory mediators. Specifically, the results demonstrated that higher levels of LXA4 were linked to a reduced obesity risk, with each unit increase corresponding to a 0.926-fold decrease in the likelihood of obesity. Conversely, elevated levels of TNF-? and VEGFR1 were associated with an increased risk of obesity. Conclusion: The study concluded that increased body mass index during pregnancy affects the levels of plasma lipoxin, cytokines, and angiogenesis-related factors. Although the exact mechanisms remain unclear, the observed changes suggest a disruption in the metabolic systems of women with obesity, which may influence physiological changes during pregnancy and lead to obesity-related pathological conditions.

    Osteoarthritis (OA) is the most commonchronic joint disease, primarily due to aging.Chondroitin sulfate (CS) is a glycosaminoglycan(GAG) commonly used to treat osteoarthritis. CS candelay the progression of pathology or reversemorphological changes in joint structure.Traditionally CS is produced from animal sources.However, due to different reasons such ascontamination, ecological risA, and the possibility ofinfectious diseases, the trend towards microbialsources has increased because of its advantages suchas purer, more antiallergic, and lower MolecularWeight (MW) than animal sources. Biochemicalanalysis of Microbial CS (MCS), which is a new andsignificant alternative as a source of CS in the OAhealing process, has not been evaluated in theliterature yet. This study was designed to analyze thebiochemical effects of MCS produced by our teamfrom a microbial source, with an MW value of 269Daltons, on the osteoarthritis healing processcompared to the commercial form. We aim to reachdata that MCS has a higher antioxidant effect thananimalYsourced CS, and in this way, it is a moresuitable production for the treatment ofosteoarthritis. In this study, Anee osteoarthritis wassurgically induced in experimental rabbits; and TGFY1?, CAT, MPO, TOS, and OSI parameters measuredin blood samples before the operation and after thehealing period were analyzed comparatively. Afterthe surgical application, the rabbits were randomlydivided into three groups: control, animalYsourcedCS, and E. coli sourced MCS. The standard rabbitdiet was administered daily to 10 rabbits in Group 1(control), and CS and MCS were applied daily to theother groups as 17 mg/Ag for 12 weeAs. Bloodsamples were taAen from rabbits at the 12th weeAafter surgery, and TGFY1?, CAT, MPO, TOS, andOSI parameters were biochemically evaluated. Thisstudy has confirmed that the antioxidant propertiesof MCS and data on its effectiveness in controllingoxidative stress compared to animalYsourced CS.Based on these results, it can be concluded that MCShas a significant potency of nutraceutical andtherapeutic agents for OA treatment.

    Could ELABELA be a Protective Biomarker in Patients with Abnormal Uterine Bleeding?

    Aim: Abnormal uterine bleeding (AUB) is a health problem characterized by various symptoms such as heavy and prolonged menstrual bleeding, affecting approximately 30% of female patients both physiologically and psychologically. The objective of this study was to assess serum Elabela (ELA) concentrations in women aged 18 and above diagnosed with functional AUB, and to compare these concentrations with those of healthy women. Material and Method: This prospective case-control study was performed from August 18, 2022 to December 30, 2022. This was a cross-sectional study including 50 women who applied to the gynecology service of Malatya Turgut Özal Training and Research Hospital with complaints of AUB and 50 women without AUB who underwent gynecological examination. The presence of AUB in patients was determined based on clinical examination conducted by a gynecologist and medical records. Demographic and clinical characteristics were recorded. Serum ELA levels were determined by commercial ELISA kit. Results: Serum ELA levels was significantly lower in patients with AUB (581.54±272.25 pg/mL) compared to the healthy group (744.55±300.31 pg/mL, p=0.005). In this study, ELA in patients with AUB showed 98% sensitivity and 80% specificity with a cut off value of 411.41 pg/mL (area under the curve [AUC], 68.1%; p=0.002). Conclusion: Serum ELA levels in patients with AUB were significantly lower than in healthy women. These results show that ELA is a good predictor of the pathophysiological process of AUB.

    CRP and LDH Levels Can Be Used for Support the COVID-19 Diagnose in Intensive Care Unit Patients

    Aim: The coronavirus disease (COVID-19) has been a public health problem that causes severe acute respiratory syndrome affected all over the word since 2019. The most commonly used parameters as inflammatory response in the clinic are leukocytes, neutrophils, erythrocyte amount and serum C-reactive protein (CRP). In recent years, it has been reported that serum PCT (procalcitonin) level may be useful in the diagnosis of bacterial and viral infections. The aim of our study is to compare blood parameters that may play a supportive role to diagnose of COVID-19 in healthy control and critically COVID-19 patient groups. Material and Methods: This retrospective research was carried out in Malatya Turgut Ozal University Training and Research Hospital, Malatya, Türkiye. Total 88 critically ill patients and 90 healthy people accepted to the study and electronic medical records of patients and control group has been collected from hospital information system (HIS). COVID-19 diagnose has been confirmed by real-time polymerase chain reaction (RT-PCR) results. Results: No statistically significant difference was found between the patient and control groups according to gender in the participants included in the study. A statistically significant increase was observed in CRP, LDH, PCT, D-dimer, urea, sediment, lympocyte and neutrophil levels in COVID-19 patients. According to logistic regression analysis CRP, LDH and sediment values were found to be statistically effective in estimating the COVID-19 infection. These results also supported by ROC analysis, CRP, neutrophil, LDH, PCT and D-dimer results were determined to be distinguishing parameters for COVID-19 patients. Conclusion: We found that CRP, PCT and LDH levels higher in the COVID-19 patients and these parameters can be used to diagnose and estimate the prognose of COVID-19 infection in intensive care patients.

    Does the high sulfur content in apricots affect oxidative stress? Running title: Effect of sulfur amount on oxidative stress

    Apricots are one of the most important fruits that can be produced worldwide. Most of the importer countries are sensitive about sulfur which is used in the protection of dried apricots. The objective of this study was to compare the effects of feeding with high and low sulfur content apricots on oxidative stress parameters of the liver. In total, 42 Wistar albino rats were fed for 12 weeks with the use of dried apricots containing different amounts of sulfur and sun-dried apricots. There was no significant difference in IL-1?, 8-OHdG and TOS levels between the control group and the group fed with sun dried apricots. There was no statistically significant difference between the groups fed with low and high sulfur-containing apricots.Non-necrotic changes were observed in 3000 ppm and 4000ppm groups in microscopic examination. No significant effect of feeding with sulfur treated apricots was found in the liver tissues of rats.

    Effect of Altered Iron Metabolism on Hyperinflammation and Coagulopathy in Patients with Critical COVID-19: A Retrospective Study

    Abstract: A novel coronavirus disease 2019 (COVID-19) outbreak has started in Wuhan, China, caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). The relationship between altered iron homeostasis and hyperinflammation may be hallmarks of COVID-19 disease. We aimed to compare some iron (ferritin and iron), inflammation (C-reactive protein [CRP], hemoglobin, lactate dehydrogenase [LDH], neutrophil) and coagulation (prothrombin time [PT], activated partial thromboplastin time [APTT], D-dimer, platelet) marker results of critical COVID-19 patients with healthy controls results. In this single center retrospective study, 50 critical patients diagnosed with COVID-19 were included, demographic, clinical characteristics, severity of disease and laboratory test results were elicited from electronic medical records and compared to 50 healthy people. A statistically significant increase in CRP, LDH, neutrophil, PT, APTT, D-dimer ferritin levels was observed in critical COVID-19 patients compared with healthy people while a statistically significant decrease was observed in hemoglobin and iron levels. In addition, no statistically significant change in platelet levels was observed. Ferroptosis may be a significant cause of multiple organ failure in critical COVID-19 patients. Ferroptosis inhibitors might have potential to combat ferroptosis in COVID-19. Therefore, larger studies are needed to ferroptosis in COVID-19 in vivo and in vitro.

    Investigation of changes in Hematological parameters and levels of Oxidative stress factors in castrated Cats

    This study was carried out to determine the effect of castration procedure on hemogram and leukogram values and the levels of oxidative stress factors in serum tissue in cats. Preoperative and postoperative 10th day hemogram/leukogram values and total oxidant/antioxidant capacities of 19 castrated cats were evaluated in the study. When the hemogram and leukogram values were examined, it was determined that there was a statistically significant increase in the amount of eosinophils (P<0.05). Although the increase in white blood cells (WBC), neutrophil and lymphocyte levels and the decrease in red blood cells (RBC) and hematocrit (HCT) levels were remarkable, it was not found to be statistically significant (P>0.05). When the levels of oxidative stress factors in the serum tissues of the cats were examined, it was determined that there was a decrease in the total antioxidant status (TAS) and an increase in the total oxidant status (TOS), but the difference in the results was not statistically significant (P>0.05). In conclusion, in this study, it was determined that the castration procedure in cats did not cause a significant difference in hemogram and leukogram parameters and the level of oxidative stress factors in serum tissue, except for the amount of eosinophils.

    Investigation of the Effects of Sun-Dried Apricot with Different Sulfur Levels on Oxidative Stress Markers

    Objective: Apricots are mostly consumed in dried form worldwide. Studies are reporting some allergic effects of dried apricot because of its content of sulfur, which is used in the drying process. Therefore sulphur content has been limited by many countries. This study was conducted to determine the effects of sun-dried apricots with different sulfur contents on nitric oxide (NO), malondialdehyde (MDA) and glutathione (GSH) that are among the markers of oxidative stress. Methods: A total of 84 Wistar albino rats were divided into groups as control, sun-dried apricot, 1000, 2000, 3000 and 4000 ppm sulfured groups. At the end of the 12th week, rats were sacrificed under general anesthesia. NO, MDA and GSH levels were studied at the end of the experiment from the ovarian and testicular tissues of the rats. Results: No statistically significant differences were found among the groups in terms of NO, MDA and GSH in the ovarian and testicular tissues removed from the rats Conclusion: We claim that sulfur amount of dried apricots is not affect on reproductive system. This study is guiding further studies that will be conducted against the restrictions in sulfur used in the drying process of dried apricots, which is economically very important for dried fruits sector.

    Koranavirüs Hastalığının Önlenmesi ve Tedavisi İçin Şifalı Bitkiler

    2019-nCoV (yeni Korona Virüsü) salgını Uluslararası Acil Halk Sağlığı Kuruluşu olan Dünya Sağlık Örgütü (WHO) tarafından 30 Ocak 2020'de ilan edildi ve DSÖ 12 Şubat'ta, 2019-nCoV patojenini SARS-CoV-2 ve Corona virüs Hastalığı 2019 (COVID-2019) olarak adlandırdı. COVID-19 11 Mart 2020’de resmi olarak WHO tarafından Pandemi olarak kabul edildi. Bitkiler tüm dünyada, viral hastalıklar da dahil olmak üzere birçok kronik enfeksiyonu tedavi etmek için yüzyıllardır geleneksel ilaç olarak kullanılmaktadır. Son yıllarda, bilim adamları fonksiyonel ve nutrasötik gıdalar üzerinde yaptıkları araştırmalarla fonksiyonel bileşiklerin insan sağlığını koruma ve hastalıkları iyileştirme potansiyelini doğrulamaya çalışmaktadırlar. Geleneksel tıbbi bitkiler, sağlıklı yaşamın sürdürülmesi, günlük hayatta alınan toksinler, hastalıklarla mücadele ve önleme, uzun ömür gibi birçok araştırma ile desteklenmiş uzun bir geçmişe sahiptir. Etnomedikal bitkiler ve doğal fitokimyasalların antiviral, antiinflamatuar ve antioksidan özellikleri üzerine yapılan araştırmalar, Covid-19 tedavisinin yanısıra çeşitli rahatsızlıklara karşı büyük bir potansiyel ilaç kaynağı olarak kabul edilebilir. Bu çalışmadan yola çıkarak bitki özütlerinin Covid-19 gibi ölümcül sonuçlara yol açabilen viral enfeksiyonlarda beyaz kan hücrelerinin ve lenfositlerin sayısındaki yükselişle bağışıklığa arttırma, proinflamatuar sitokinlerin üretimini ve salımını düzenleme, C-reaktif protein ve eritrosit sedimantasyon oranındaki düşüşle anti-inflamatuar bir etki gösterme, virüsün gelişimine müdahale etme gibi olumlu etki ve potansiyel antiviral ajan aktivitesi sergilediği görülmektedir. Bu çalışmada COVID-19 enfeksiyonu ile ilişkili fitokimyasallar ve etkileri derlenmiştir

    Kuru kayısılardaki kükürt miktarının serum oksidatif stres parametleri üzerine etkisi

    Kayısı, dünyanın farklı bölgelerinde yetiştirilebilen ve ticari öneme sahip bir meyvedir. Kayısılar güneşte veya depolanma süresini uzatmak amacıyla kükürtle muamele edilerek kurutulmaktadır. Özellikle Avrupa Birliği ülkeleri ve ülkemizde kuru kayısıda bulunması gereken kükürt miktarı 2000 mg/L olarak belirlenmiştir ve bu miktar azaltılmak istenmektedir. Çalışmanın amacı, yüksek ve düşük kükürt içerikli kayısılarla beslenmenin serum oksidatif stres parametreleri üzerine etkilerini karşılaştırmaktır. Toplamda 84 adet Wistar albino rat, gün kurusu kayısı ve farklı miktarda kükürt içeren kuru kayısıların kullanıldığı yemlerle 12 hafta boyunca beslendi. Ratların serum Total antioksidan seviye, Total oksidan seviye, Malondialdehit ve Glutatyon seviyeleri ölçüldü. En yüksek total antioksidan seviye gün kurusu kayısı ile beslenen grupta, en düşük total antioksidan seviye ise 3000 mg/L grubunda gözlemlenmiştir. Malondialdehit seviyelerinde sadece 1000 mg/L grubunda, Glutatyon seviyelerinde ise 1000 mg/L, 2000 mg/L ve 3000 mg/L gruplarında anlamlı artışlar görülmüştür.

    The relationship between dietary antioxidant capacity and serum oxidative stress index of hemodialysis patients

    Purpose: Oxidative stress increases in uremic conditions such as kidney failure and during hemodialysis. In this study, it was aimed to examine the relationship between the total antioxidant capacity of the diet and the serum oxidative stress (sOSI) index in hemodialysis patients. Materials and Methods: In this study conducted with 82 individuals (41 patients and 41 controls). Demographic characteristics and anthropometric measurements of the individuals were taken by researchers. In order to determine the nutritional status of the individuals, 3-day food consumption records were taken and the total antioxidant capacity of the diet was calculated. Results: The mean age of the individuals participating in the study was found to be 54.01±2.24 years. Hemodialysis treatment mean for 4.8±3.12 years. While 41.46% of hemodialysis patients were moderately malnourished. Dietary Oxygen Radical Absorption Capacity (dORAC) (?mol) measurement result was found to be 2415.3±1073.45, 5468.4±2393.85 in the hemodialysis and control groups, respectively. In the multiple logistic regression analysis performed, the decrease in body mass index (OR 2.21 95% CI 1.050-2.317) and triceps skinfold thickness values (OR 3.22 95% CI 1.722-4.001) in hemodialysis patients led to an increase in the sOSI value. It has been observed that when dietary protein (OR 1.23 95% CI 1.003-3.189), vitamin C (OR 2.88 95% CI 1.004-3.106) and dTAC (OR 2.04 95% CI 1.967-2.001) increase, the sOSI value decreases in hemodialysis patients. Conclusion: There is a positive relationship between diet total antioxidant capacity and serum antioxidant level, and it was determined that the dietary antioxidant capacity of hemodialysis patients was significantly lower than that of healthy individuals. According to the results of the study, it is recommended to monitor the food consumption status of hemodialysis patients in order to increase the dietary antioxidant capacity.
